RASV-inoculation prevents cancer and metastasis IRF1-driven IL-7 expression in bone marrow through CD8+ T-cell dependent manner
Despite advances in cancer treatment, tumor recurrence remains a major clinical challenge and is closely associated with poor long-term outcomes. Residual tumor cells and insufficient durable antitumor immunity can contribute to disease relapse even after an initial therapeutic response. Therefore, strategies that establish long-lasting immune surveillance are required to effectively prevent tumor recurrence. Thus, inducing systemin and durable anti-cancer immune responses of host has emerged as a strategy to prevent cancer metastasis. Herein, we investigated cancer prevention strategy by reprogramming hematopoietic stem cells by inoculating Recombinant-attenuated Salmonella vaccine (RASV), analogous to BCG vaccine model. RASV pre-inoculation inhibited tumor growth in CD8 T cell-dependent manner. We observed changed hematopoietic stem cell population in bone marrow (BM) which contributes to anti-cancer effects. IL-7-induced common lymphoid progenitors (CLPs) were significantly affected by RASV inoculation. IL-7 was expressed by IFN-γ-induced IRF1, and IL-7R blocking curtailed RASV-induced anti-cancer effects. Finally, RASV inoculation prevented metastasis of 4T1, which is a highly metastatic triple negative breast cancer, in mouse lung. This study suggests a critical role of HSCs reprogramming in BM for preventing cancer metastasis highlighting the IFN-γ-IRF1-IL-7 axis in BM.
