One-pot ​three-component coupling approach to indolizine-pyrrolodiazepine hybrids

A new indolizine-pyrrolodiazepine hybrid skeleton was readily constructed by way of one-pot three-component coupling of pyridine-2-acetonitrile, N-substituted pyrrole-2-carboxaldehyde, and TMSCN in the presence of DBU where a domino aldol condensation-Michael addition-cycloisomerization-imine formation sequence allowed for construction of two central rings (pyrrole and diazepine) of the tetracyclic product with formation of two C-C and two C-N bonds.
